Triclabendazole is a benzimidazole-derived flukicide with a comparatively narrow parasite spectrum. Its distinctive clinical role is activity against susceptible early immature, later immature and adult Fasciola stages. This stage coverage supports treatment of acute as well as chronic fasciolosis in ruminants, depending on product and parasite susceptibility.
Group
Benzimidazole flukicide
Mechanism of Action
Triclabendazole is converted to active sulfoxide and sulfone metabolites. These interfere with fluke microtubule-associated transport, protein handling and tegumental function, producing structural and metabolic damage. The resulting disruption affects developing and adult susceptible flukes. Its complete molecular mechanism differs in detail from that of conventional nematode-targeting benzimidazoles and remains incompletely characterized.
Dose Rates
Cattle
| Use / stage | Dose | Route | Schedule | Instructions |
|---|---|---|---|---|
| Fasciolosis | 12 mg/kg | PO | Once |
Sheep
| Use / stage | Dose | Route | Schedule | Instructions |
|---|---|---|---|---|
| Fasciolosis | 10 mg/kg | PO | Once |
Indications
- Treatment of acute, subacute or chronic fasciolosis due to susceptible Fasciola hepatica in sheep or cattle with an appropriate product.
- Treatment of susceptible F. gigantica infection in supported ruminant species, with host- and parasite-specific evidence.
Contraindications
- Hypersensitivity; food-animal milk restrictions vary by exact product.
Interactions
- Do not substitute combination products without checking all active ingredients.
Adverse effects
- Adverse reactions are uncommon at recommended doses; hypersensitivity is possible.
Toxicity
Marked overdose may cause reduced appetite, weight loss and altered activity; treat supportively.
Preparations
| Preparation | Form | Strength | Route |
|---|---|---|---|
| Tribex 100 mg/mL oral suspension | Oral Liquid | 100 mg / mL | PO |
| Olgard oral suspension Also contains ivermectin 0.1% w/v; the single-drug regimen does not determine safe combination use. | Oral Liquid | triclabendazole 50 mg / mL + ivermectin 1 mg / mL | PO |
Monitoring and Administration
Shake suspension, weigh animals accurately and investigate suspected resistance rather than repeatedly redosing.
Tribex 100 mg/mL: cattle meat withdrawal 56 days; not for lactating animals producing milk for consumption. Dry-cow use has specific calving restrictions.
